Von Willebrand Factor API Manufacturer and Supplier in India

Von Willebrand Factor (VWF) is a large multimeric glycoprotein that plays an essential role in primary haemostasis and blood coagulation. It promotes platelet adhesion at sites of vascular injury by interacting with exposed subendothelial collagen and platelet glycoprotein receptors. VWF also functions as a carrier and stabilizing protein for coagulation Factor VIII in the circulation. Human VWF is encoded by the VWF gene and consists of 2,813 amino acids in its precursor protein.

Von Willebrand Factor is an important biological active substance used in the development and manufacture of therapies for von Willebrand disease (VWD) and related bleeding disorders. Depending on the manufacturing platform, VWF can be obtained from human plasma or produced as recombinant human von Willebrand Factor. Recombinant VWF, such as vonicog alfa, is produced using recombinant DNA technology in Chinese hamster ovary (CHO) cells.

The structure of VWF is substantially different from that of conventional pharmaceutical small molecules. It exists as multimers with molecular sizes ranging from approximately 500 kDa to more than 20,000 kDa. Its biological activity depends on its molecular structure, multimer distribution, glycosylation, and functional interaction with platelets, collagen and Factor VIII.

VWF contains several functional domains that contribute to its biological activity. The D′D3 region participates in Factor VIII binding, while the A1 domain interacts with platelet glycoprotein Ib and other ligands. The A3 domain contributes to collagen binding. These structural characteristics are important when evaluating the identity, purity and functional performance of a VWF biological substance.

For pharmaceutical applications, VWF is not appropriately characterized using only conventional small-molecule tests such as melting point, residue on ignition or standard HPLC assay percentage. Instead, its quality assessment relies on protein identity, purity, molecular characterization, VWF functional activity, antigen content, multimeric distribution and other biological attributes.

A key functional measurement for VWF is VWF:RCo activity, historically known as ristocetin cofactor activity. Pharmaceutical VWF products express potency in International Units (IU), with VWF:RCo activity measured against an appropriate international standard. For example, the recombinant VWF product vonicog alfa is labelled according to VWF:RCo potency, and its reported specific activity is approximately 110 IU VWF:RCo/mg protein.

VWF pharmaceutical products may also be evaluated for VWF antigen (VWF:Ag), collagen-binding activity, Factor VIII binding, multimeric distribution, protein purity, aggregates and degradation products. These tests provide a more meaningful assessment of a biological VWF substance than conventional chemical purity measurements.

Von Willebrand Factor is particularly relevant to therapies designed for patients with von Willebrand disease, an inherited bleeding disorder associated with quantitative or qualitative defects of VWF. VWF replacement products are used to restore the functional activity required for normal platelet adhesion and to support Factor VIII stability.

Recombinant VWF offers a defined manufacturing platform and is produced using controlled cell-culture and purification processes. Regulatory assessment of recombinant VWF includes characterization of protein structure, functional activity, purity, multimer distribution and manufacturing controls.

Swapnroop Drugs & Pharmaceuticals supports pharmaceutical and biopharmaceutical requirements for Von Willebrand Factor API and biological active substances. VWF-related materials can be considered for research, development, formulation and pharmaceutical manufacturing applications, subject to applicable regulatory and product-specific requirements.

For pharmaceutical development, the appropriate VWF specification should be established according to the source of VWF, manufacturing platform, dosage form and applicable pharmacopoeial or regulatory requirements. Plasma-derived and recombinant VWF products can have different analytical profiles and labelled potency specifications.

Von Willebrand Factor is therefore best described as a biological protein API / coagulation factor rather than a conventional chemical API. Its quality is determined through a combination of physicochemical characterization and biological functional testing to ensure identity, purity, potency and consistency.
