Sorbitol DC Granules

Sorbitol DC Granules are pharmaceutical excipient granules based on d-sorbitol and designed in a physical form suitable for direct-compression formulation development. Sorbitol, also known as d-glucitol, has the molecular formula C₆H₁₄O₆, molecular weight 182.17, and CAS Number 50-70-4. The current USP-NF monograph defines Sorbitol as containing 91.0% to 100.5% d-sorbitol, calculated on the anhydrous basis.

The designation DC, or Direct Compression, refers to the intended processing application of the finished granulated material. It does not mean that every sorbitol material is inherently suitable for direct compression. The suitability of a particular Sorbitol DC Granules grade depends on its particle-size distribution, flowability, bulk density, compressibility, compactability, moisture level, and other validated physical properties.

What Are Sorbitol DC Granules?

Sorbitol DC Granules are granulated sorbitol excipient materials developed for pharmaceutical manufacturing processes in which the material may be incorporated directly into a powder blend and subsequently compressed into tablets, subject to formulation suitability.

Direct compression requires excipients with appropriate flow and compaction characteristics. A controlled granulated form can provide more consistent particle characteristics than an uncontrolled fine powder and may support improved material handling, blending, die filling, and compression.

The exact performance of Sorbitol DC Granules is dependent on the manufacturing process and finished-product specification. Therefore, properties such as particle size, bulk density, tapped density, flowability, compressibility, compactability, and moisture should be controlled according to actual product data.

Sorbitol as a Pharmaceutical Excipient

Sorbitol is a polyhydric alcohol used as a pharmaceutical excipient. Current USP-NF identifies Sorbitol as d-glucitol and specifies a d-sorbitol content of 91.0% to 100.5% on an anhydrous basis. Sorbitol has the formula C₆H₁₄O₆ and molecular weight 182.17.

Sorbitol is also highly water soluble. FCC describes sorbitol as very soluble in water and notes that it occurs in forms including powder and granules. These characteristics can make sorbitol useful in formulation systems where a water-soluble polyol excipient is required.

For direct-compression applications, however, chemical identity and solubility alone are not sufficient to establish suitability. Physical properties of the finished DC grade are important for manufacturing performance.

Direct Compression Characteristics

Direct compression is a manufacturing approach in which a powder or granulated blend is compressed without a separate wet- or dry-granulation step immediately before tableting.

For Sorbitol DC Granules, important formulation and processing attributes can include:

  • Particle-size distribution
  • D10, D50 and D90
  • Bulk density
  • Tapped density
  • Flowability
  • Compressibility
  • Compactability
  • Fines content
  • Moisture content
  • Granule strength
  • Segregation behavior

These parameters can affect powder movement, die filling, tablet weight consistency, compression behavior, and finished tablet properties.

Particle Size Distribution

Particle size is an important consideration for direct-compression excipients. A controlled particle-size distribution can support consistent flow and reduce excessive segregation during blending.

Sorbitol DC Granules may be evaluated using parameters such as D10, D50 and D90. These values describe different points of the particle-size distribution and can be used during product development and quality control.

The actual particle-size limits should be taken from the approved product specification. Universal D10, D50 or D90 values should not be assigned without product-specific data.

Bulk Density and Tapped Density

Bulk density and tapped density are useful physical parameters for evaluating granulated excipients.

Bulk density describes the mass of material per unit volume before standardized tapping, while tapped density reflects the packing behavior after a defined tapping procedure. These values can influence hopper filling, die filling, blend volume, packaging, and tablet manufacturing.

The relationship between bulk and tapped density can also be used to calculate derived flow indicators such as Carr Index and Hausner Ratio when the required measurements are available.

Flowability

Flowability is particularly important for direct-compression materials because the excipient must move consistently through manufacturing equipment and fill the tablet die uniformly.

Flow performance can be influenced by:

  • Particle size
  • Particle shape
  • Fines content
  • Surface characteristics
  • Moisture
  • Bulk density
  • Granule strength
  • Processing conditions

A well-controlled Sorbitol DC Granules grade should therefore be evaluated using appropriate flow testing rather than assuming a universal flow classification.

Compressibility and Compactability

Compressibility and compactability are important characteristics when a granulated excipient is intended for tablet manufacture.

Compressibility describes how a powder bed changes in volume under applied pressure, whereas compactability relates to the strength of the resulting compact at a given compression pressure.

For Sorbitol DC Granules, these properties should be evaluated experimentally during formulation development. Tablet hardness, tensile strength, friability, disintegration, and dissolution can also be influenced by formulation composition and compression conditions.

No universal tablet hardness or compression-force value should be assigned to Sorbitol DC Granules without finished formulation data.

Moisture and Hygroscopicity

Sorbitol is hygroscopic, so moisture management is an important consideration during the manufacture, packaging, and storage of Sorbitol DC Granules.

Moisture can affect powder flow, particle cohesion, granule strength, compaction, and stability. Excessive moisture exposure may also influence handling characteristics during blending and compression.
