Clodronic Acid API

Clodronic Acid, also known as Clodronate, is a first-generation bisphosphonate active pharmaceutical ingredient belonging to the class of non-nitrogen-containing bisphosphonates. It is chemically characterized by a dichloromethylene group attached to two phosphonic acid groups.

Clodronic Acid has the molecular formula CH₄Cl₂O₆P₂, molecular weight 244.89 g/mol, and CAS number 10596-23-3. It is an achiral compound with no defined stereocenters or optical activity.

Pharmaceutical Applications

Clodronic Acid and its pharmaceutical salts are associated with inhibition of bone resorption and reduction of abnormal calcium release from bone. Clodronate pharmaceutical preparations have been used in the management of conditions involving increased bone resorption, including hypercalcaemia associated with malignancy and osteolytic bone disease.

The API is particularly relevant to pharmaceutical formulations containing clodronate, with disodium clodronate being an important pharmaceutical salt form.

Key Product Features

  • Bisphosphonate API
  • Non-nitrogen-containing bisphosphonate
  • Bone-resorption inhibitor
  • Contains two phosphonic acid groups
  • Achiral chemical structure
  • No defined stereocenters
  • Suitable for pharmaceutical formulation development
  • Available as parent acid or as pharmaceutical salt forms, depending on product specification

Mechanism of Action

Clodronate belongs to the bisphosphonate class. Bisphosphonates have a strong affinity for bone mineral and become associated with areas of active bone remodeling.

After uptake by bone-resorbing cells, clodronate can interfere with cellular energy metabolism and promote osteoclast dysfunction and apoptosis. This contributes to suppression of excessive bone resorption.

Its pharmacological activity is therefore associated with:

  • Reduction of osteoclast activity
  • Inhibition of bone resorption
  • Reduction of excessive calcium release from bone
  • Modulation of pathological bone turnover

Chemical and Physical Characteristics

Clodronic Acid is a highly polar phosphonic-acid-containing compound. The parent compound is chemically represented as (Dichloromethylene)diphosphonic acid.

Clodronic Acid and Clodronate Disodium

For your website, this distinction is important.

Clodronic Acid:
CAS 10596-23-3
Formula CH₄Cl₂O₆P₂
Molecular Weight 244.89 g/mol

Clodronate Disodium:
CAS 22560-50-5
Formula CH₂Cl₂Na₂O₆P₂
Molecular Weight 288.85 g/mol

Clodronate Disodium Tetrahydrate:
Formula CH₁₀Cl₂Na₂O₁₀P₂
Molecular Weight 360.91 g/mol.

Therefore, if your company's actual product is Clodronate Disodium, do not use the parent-acid molecular weight of 244.89 g/mol on the product page.
